研究背景：CD40 配體（CD40L；CD154；TRAP）屬于腫瘤壞死因子 (TNF) 家族，是 CD40/TNFRSF5 的配體，特異性表達于活化的 CD4+ T 淋巴細胞[1]。 CD40L 是 B 細胞上的一種 II 型跨膜蛋白，可觸發 B 細胞分化、成熟和凋亡的重要信號[4]。 CD40L 通過與 T 細胞交聯來發揮作用產生共刺激信號，從而與 TCR/CD3 連接和 CD28 共刺激一起增強 IL4 和 IL10 的產生，并促進干擾素-γ 和 TNF-α[1] 的產生[4]。 CD40L 與抗原呈遞細胞 (APC) 上的 CD40 結合，通過刺激 I-κB 激酶 (IKK) 激活 TNFR 相關因子 2 和 IKK2 依賴性通路，增加 NF-κB DNA 結合和 p65 核轉位。I-κB 激酶的激活導致 c-Jun N 末端激酶強烈激活以及 GST-I-κB 和 GST-p65 磷酸化[2]。 CD40L 參與 MAPK強烈抑制 Bcl-6 的途徑，誘導 Erk1/2、p38 和 Jnk1/2 磷酸化并激活 NF-κB[3] 介導的 IRF4。 CD40L 還結合并發出信號通過幾個整合素，包括 αvβ3 和 α5β1，它們與 CD40L 的三聚體界面結合。CD40L 在免疫反應中發揮重要作用，是炎癥的主要靶點[5]。 CD40L 廣泛存在于不同動物中，而人類中的序列與恒河猴高度相似（ 98.08%），但與大鼠和小鼠有很大不同，相似度分別為 77.31% 和 77.69%。人體內的CD40L被切割成膜形式(1-261aa)和可溶性形式(113-261aa)的2條鏈，而可溶性形式是通過蛋白水解加工從膜形式衍生出來的。血小板中可溶性 CD40L 的釋放部分受到 GP IIb/IIIa、肌動蛋白聚合和基質金屬蛋白酶 (MMP) 抑制劑敏感途徑[6]的調節。
