鉆入皮膚的幼蟲在皮下移行進入皿管或淋巴管，進而被帶入右心，經過肺動脈進入肺血管，大部分幼蟲可以繼續穿過微血管到達肺泡。沿支氣管、氣管，再隨宿主的吞咽動作經食管、胃到達小腸，一部分幼蟲也可隨痰被吐出。幼蟲到達小腸后，迅速成長，感染3～4天后開始第三次蛻皮，成為第四期幼蟲，1 0天后進行第四次蛻皮，逐漸發育為成蟲。自幼蟲鉆入至成蟲交配產卵，一般需要5~7周時間。[2] 兩種鉤蟲的致病作用相似。十二指腸鉤蚴引起皮炎者較多，成蟲導致的貧血亦較嚴重，同時還是引起嬰兒鉤蟲病的主要蟲種，因此，十二指腸鉤蟲較美洲鉤蟲對人體的危害更大。人體感染鉤蟲后是否出現臨床癥狀，除與鉤蚴侵入皮膚的數量及成蟲在小腸寄生的數量有關外，也與人體的健康狀況、營養條件及免疫力有密切關系。有的雖在糞便中檢獲蟲卵，但無任何臨床癥象者，稱為鉤蟲感染（hookworm infection）。

Some people detect eggs in the feces, but no clinical signs are called hookworm infections.
